What is this medication?

LYSODREN (Mitotane) is indicated for treatment of adrenocortical carcinoma.

ALERT: WARNING
In patients taking mitotane, adrenal crisis occurs in the setting of shock or severe trauma and response to shock is impaired. Administer hydrocortisone, monitor for escalating signs of shock, and discontinue mitotane until recovery occurs.

How does this medication work?

LYSODREN (Mitotane) is an adrenolytic agent that directly suppresses the adrenal cortex and alters the metabolism of steroids in the periphery.

How should I take this medication? 

Oral: Take 2 to 6 g per day in 3 to 4 divided doses initially, then increase incrementally to achieve a blood concentration of 14 to 20 mcg/mL or as tolerated.

How to Take: Follow instructions provided by physician/pharmacy label, take consistently at same times each day unless otherwise directed. Mitotane is associated with a moderate emetic potential so antiemetics may be needed to prevent nausea and vomiting.

You Need to Avoid:  Significant drug interactions exist, requiring dose/frequency adjustment or avoidance so avoid starting any new medications without consulting prescriber.

What should I watch for while using this medication? 

Before starting LYSODREN (Mitotane), make sure your physician is aware of any allergies or medications you currently take, if you have hepatic impairment, are pregnant, or breastfeeding. Mitotane use has been associated with rare but serious adverse events such as adrenal crisis, adrenal insufficiency, CNS toxicity, gastrointestinal toxicity, ovarian macrocysts, prolonged bleeding time, and thyroid dysfunction.

What if I miss a dose?

If you miss a dose of medication, try to take it as soon as possible. However, if it is almost time for your next dose, take only that scheduled dose. Do not take double or extra doses.

How should I store this medication?

Keep out of the reach of children at all times. Store at room temperature, 59 to 86° F (15 to 30° C).  Protect from light. Keep the container tightly closed. Properly dispose of any unused medication after the expiration date.

What are the possible side effects of using this medication? 

Skin rash, anorexia, diarrhea, nausea, vomiting, depression, dizziness, vertigo.
